Wheat stripe rust, caused by the fungal pathogen Puccinia striiformis f.sp. tritici (PST), is a major disease of wheat in temperate‐cold climates. The identification of new markers would ease the procedure for evaluating the ongoing pathogen evolution. Twelve single pustule isolates were generated from samples of PST obtained in UK during 1987–2001. They were evaluated for their pathogenic behaviour on a set of differential cultivars and were analysed by sequence‐related amplified polymorphisms (SRAP) technique, to identify polymorphisms useful to evaluate variability among isolates. This is the first report of the application of SRAP technique to Uredinales order.  相似文献

四川省小麦条锈菌群体遗传多样性的SSR分析   总被引:1,自引:0,他引:1  
利用TP-M13-SSR自动荧光检测技术,对四川省小麦条锈菌群体遗传多样性水平进行了分析。研究结果表明,四川省小麦条锈菌群体遗传多样性比较丰富,地区之间存在明显的差异,川西北和四川盆地的种群遗传多样性相对较高,而四川西南部和四川东南部的种群遗传多样性较低。四川小麦条锈菌群体存在一定的遗传分化,地区间的遗传变异仅占14.92%,群体间的遗传变异占总变异的23.06%,群体内遗传变异占60.02%,遗传变异主要存在于群体内部。基因流和共享基因型从分子水平证实了四川小麦条锈菌在地区间的传播,且川西北和四川盆地之间的菌源交流最为广泛。  相似文献

以雷公藤(Tripterygium wilfordii Hook f.)根愈伤组织为材料,以NT为基本培养基,添加不同浓度硫代硫酸钠、抗坏血酸、柠檬酸、硝酸银、聚乙烯吡咯烷酮以及活性炭,探讨抗褐变剂对雷公藤愈伤组织生长、抗褐化和雷公藤内酯醇及总生物碱含量的影响。结果表明:除较低浓度柠檬酸以外,其他抗褐变剂均不同程度抑制雷公藤愈伤组织的生长。培养基中添加不同浓度硫代硫酸钠、抗坏血酸、柠檬酸、聚乙烯吡咯烷酮以及活性炭,雷公藤愈伤组织褐化程度不但没有得到控制,反而使褐化加重。但所有处理中,愈伤组织中内酯醇含量明显升高,其中加入50mg/L柠檬酸处理的内酯醇含量为对照的2.4倍。培养基中加入10~50mg/L硝酸银不仅能抑制雷公藤愈伤组织褐变,内酯醇的含量也随着硝酸银浓度的增加而增加。供试抗褐变剂中除较低浓度硫代硫酸钠以外,其他处理对雷公藤总生物碱的合成起抑制作用。  相似文献

Winter wheat was grown for six successive years (Expt 1) and for three successive years (Expt 2) in field experiments on different soil types. Artificial inoculum of the take-all fungus (Gaeumannomyces graminis var. tritici cultured on autoclaved oat grains) was incorporated in the soil of some of the plots just before, or at, sowing of the first winter wheat crop. Expt 1 tested the incorporation of similar amounts of inoculum (212 kg ha-1) at different depths. Expt 2 tested different amounts of inoculum at the same, shallow depth. Early sowing (September), late sowing (October) and spring inoculation were additional treatments, applied to the first crop only, in Expt 2. Seasonal factors apart, the disease outcome in the first year after inoculation depended on amounts and placement of applied inoculum, as well as date of sowing. Deeper inoculum resulted in less disease (Expt 1). Severe take-all was produced in Expt 2 by incorporating inoculum shallowly in sufficient quantities (400 kg ha-1 or more). Less inoculum (200 kg ha-1) generated less disease, especially in earlier-sown plots. Differences in disease amongst inoculum treatments were greatest in the first year and diminished subsequently, particularly where sowing had been early in the first year. In Expt 1, where first crops exposed to artificial inoculum developed moderate-to-severe disease, disease in subsequent second and/or third crops was less. In the fourth crop a second peak of disease occurred, coinciding with a first peak in sequences without added inoculum. Take-all decline (TAD) appeared to be expressed in all sequences thereafter. In Expt 2 in sequences without added inoculum, TAD occurred after a peak of disease in the second crop. Where 400 kg ha-1 or more of inoculum were added, disease was severe in the first year and decreased progressively in successive years. Disease was less patchy in plots that received artificial inoculum. However, it remains uncertain mat severe disease caused by artificial inoculation achieved an early onset of true TAD. The infectivity of the top 12 cm of soil in the first 3 yr of Expt 1, determined by bioassay, depended on the depth of added inoculum and amount of disease in subsequent crops. However, at the time of the naturally occurring peak of disease severity (in either inoculated or non-inoculated plots) it did not predict either disease or TAD. Differences and similarities amongst epidemics developing naturally and those developing from different amounts and placement of applied inoculum have been revealed. The epidemiological implications of adding inoculum and the potential value of artificially-created epidemics of take-all in field trials are discussed.  相似文献

Many reports point to the existence of a network of regulatory signalling occurring in plants during the interaction with micro-organisms (biotic stress) and abiotic stresses such as wounding. However, the focus is on shared intermediates/components and/or common molecular outputs in differently triggered signalling pathways, and not on the degree and modes of effective influence between abiotic and biotic stresses nor the range of true plant-pathogen interactions open to such influence. We report on local and systemic wound-induced protection in tomato (Solanum lycopersicum L.) to four pathogens with a range of lifestyles (Botrytis cinerea, Fusarium oxysporum f.sp. lycopersici, Phytophthora capsici and Pseudomonas syringae pv. tomato). The role of ethylene (ET) in the phenomenon and in the induction by wounding of several markers of defense was investigated by using the never-ripe tomato mutant plants impaired in ET perception. We showed that PINIIb, PR1b, PR5, PR7 and peroxidase (POD) are influenced locally and/or systemically by wounding and, with the exception of POD activity, by ET perception. We also demonstrated that ET, although not essential, is positively (B. cinerea, P. capsici) or negatively (F. oxysporum, P. syringae pv. tomato) involved not only in basal but also in wound-induced resistance to each pathogen.  相似文献

Crown rust, caused by Puccinia coronata f. sp. lolii, is one of the most important diseases of temperate forage grasses, such as ryegrasses (Lolium spp.), affecting yield and nutritional quality. Therefore, resistance to crown rust is a major goal in ryegrass breeding programmes. In a two-way pseudo-testcross population consisting of 306 Lolium multiflorum individuals, multisite field evaluations as well as alternative methods based on artificial inoculation with natural inoculate in controlled environments were used to identify QTLs controlling resistance to crown rust. Disease scores obtained from glasshouse and leaf segment test (LST) evaluations were highly correlated with scores from a multisite field assessment (r = 0.66 and 0.79, P < 0.01, respectively) and thus confirmed suitability of these methods for crown rust investigations. Moreover, QTL mapping based on a linkage map consisting of 368 amplified fragment length polymorphism (AFLP) and simple sequence repeat (SSR) markers revealed similar results across different phenotyping methods. Two major QTLs were consistently detected on linkage group (LG) 1 and LG 2, explaining up to 56% of total phenotypic variance (V p). Nevertheless, differences between position and magnitude of QTLs were observed among individual field locations and suggested the existence of specific local pathogen populations. The present study not only compared QTL results among crown rust evaluation methods and environments, but also identified molecular markers closely linked to previously undescribed QTLs for crown rust resistance in Italian ryegrass with the potential to be applied in marker-assisted forage crop breeding. Predicting how populations respond to climate change requires an understanding of whether individuals or cohorts within populations vary in their response to climate variation. We used mixed-effects models on a song sparrow (Melospiza melodia) population in British Columbia, Canada, to examine differences among females and cohorts in their average breeding date and breeding date plasticity in response to the El Niño Southern Oscillation. Climatic variables, age and population density were strong predictors of timing of breeding, but we also found considerable variation among individual females and cohorts. Within cohorts, females differed markedly in their breeding date and cohorts also differed in their average breeding date and breeding date plasticity. The plasticity of a cohort appeared to be due primarily to an interaction between the environmental conditions (climate and density) experienced at different ages rather than innate inter-cohort differences. Cohorts that expressed higher plasticity in breeding date experienced warmer El Niño springs in their second or third breeding season, suggesting that prior experience affects how well individuals responded to abnormal climatic conditions. Cohorts born into lower density populations also expressed higher plasticity in breeding date. Interactions between age, experience and environmental conditions have been reported previously for long-lived taxa. Our current results indicate that similar effects operate in a short-lived, temperate songbird.  相似文献
